Their ED50 values were 0.04, 0.03, and 2 mgkg respectively. The potency of both the agonists to induce, and the antagonists to inhibit, PE was found to be dependent on their selectivity for the 5-HTlc receptor versus the 5-HT2 receptor. Spiperone 0.1 - 1.0 mgkg ; and GR 38032F 1 - 10 mgkg ; did not antagonise mCPPinduced PE. 8-OH-DPAT and 5-MeODMT counteracted mCPP 0.46 m a g ; -induced PE. Their EDs0 values were 0.03 and 0.4 mgkg respectively. DO1 counteracted mCPP-induced PE only at doses above 1 mgkg whereas CGS 12066B 1.0 - 10 mgkg ; was inactive. The results suggest that PE are induced by activation of the 5-HTlc receptor and are functionally inhibited by activation of 5-HTIAor 5-HT2 receptors. Circulating endothelial cells CECs ; may be associated with the severity and extent of atherosclerosis. We measured endothelial cells in stable RTRs. Methods Non traumatic venepuncture was performed on 25 normal subjects and 95 RTRs attending the clinic. CECs were isolated by from peripheral blood with immunomagnetic anti-CD146 a marker exclusive to human CECs ; coated Dynabeads followed by washing with phosphate buffer solution. Cells and beads were counted by light microscopy. Results Levels of CECs were significantly higher in RTRs than controls 606 [25, 3033], median [range] vs. 175 [0, 500] cells 100ml ; . 81 patients were on calcineurin inhibitor CNI ; based immunosuppression 48 cyclosporin based, 33 tacrolimus based ; , with 14 patients on CNI free regimes.
